I am as incapable of selecting one favorite fictional lesbian as any other single shot, but the first contenders look like the ironclad classics of Florian del Guiz in Mary Gentle's Ash: A Secret History (2000),

I feel like I'm overlooking earlier ones, but Alice Jones or Tilly from OUaT's S7 is the main one of mine that comes to my mind right now, although tbf, she is a good one.
